When I load it there is no duration anymore. Why?

the time duration of a route visible during the planning process is only a raw estimate based on the data from the router so it is not saved. The app offers a more accurate estimate when you start navigating the route - more parameters are considered.

I can't confirm that. When I load the route (by bike, attached down), Locus shows a duration of 8:45 hours for 37 km. It should be about 3 hours. That's what the online route planner showed.
