The most common cases : if you want Locus to calculate a route from one point to another according to your means of transport and in fully mapped area (with help of some external router like BRouter, YOURS etc.), use navigation. If you want to be guided to a geocache somewhere in free terrain without mapped roads, use guidance.

Or find a gpx track and let you guide along it (with the possibility of a warning BEEP when you go off the track more than a set distance, say 100 meters. Works great for hiking along a track.

Navigation is more useful on-ways, but useless off-ways.
